Mechanism of action:
Anastrozole is a third generation non-steroidal aromatase inhibitor. It inhibits the enzyme aromatase which is responsible for converting androgens to estrogens (aromatization). In postmenopausal women, hormone receptor positive (estrogen receptor) breast cancer is stimulated and maintained by estrogens. The suppression of estrogen biosynthesis (aromatization) is easily achieved by using Anastrozole.
Pharmacokinetics:
Absorption: Rapid (Fasted state). tmax=2 hrs.
Distribution: Plasma protein binding (40%).
Metabolism: Liver via N-dealkylation, hydroxylation and glucuronidation.
Elimination: Urine and feces (85%).
Half-life: t1/2=50 hrs.
