Amoxicillin is a broad-spectrum, semi-synthetic aminopenicillin antibiotic. It exerts bactericidal activity by inhibiting bacterial cell wall synthesis through binding to penicillin-binding proteins (PBPs), leading to cell lysis. It is active against a wide range of gram-positive and gram-negative bacteria and is one of the most widely prescribed oral antibiotics worldwide.

Indications

Admox (Amoxicillin Trihydrate) is indicated for infections caused by susceptible organisms:

• Respiratory Tract Infections: Pneumonia, bronchitis, tonsillitis, pharyngitis caused by susceptible organisms
• Ear, Nose & Throat Infections: Otitis media, sinusitis
• Urinary Tract Infections: Uncomplicated UTIs caused by E. coli, Enterococcus faecalis, Staphylococcus saprophyticus
• Skin & Soft Tissue Infections
• H. pylori Eradication: As part of triple therapy regimens
• Dental Infections & Pre-procedure Prophylaxis
• Typhoid Fever (in combination or as directed)

Dosage & Administration

Adults: 250–500mg three times daily, or 500mg–1g twice daily, depending on severity of infection and physician's instructions.

Children: 25–50mg/kg/day in divided doses every 8 hours (use suspension for children). Dose based on weight and infection type.

Duration: Typically 5–14 days as directed. Complete the full prescribed course even if symptoms improve.

Administration: May be taken with or without food. Taking with food reduces GI upset.

Contraindications & Precautions

• Contraindicated in patients with known hypersensitivity to amoxicillin, penicillins, or cephalosporins
• Use with caution in patients with renal impairment — dose adjustment required for severe impairment
• May reduce efficacy of oral contraceptives
• Caution in patients with history of allergic disorders, asthma, or urticaria
• Monitor for C. difficile-associated diarrhoea with prolonged use

Side Effects

Common: Nausea, diarrhoea, vomiting, skin rash. Hypersensitivity reactions including urticaria and maculopapular rash. Serious but rare: anaphylaxis, Stevens-Johnson syndrome, C. difficile colitis, hepatitis.

Drug Interactions

Warfarin — anticoagulant effect may be increased. Methotrexate levels may increase. Allopurinol increases risk of rash. Probenecid reduces renal excretion (increases amoxicillin levels). May reduce efficacy of live bacterial vaccines.

Storage

Store below 25°C in a dry place away from direct sunlight and moisture. Keep out of reach of children.
