Overview
Angilock Plus 50 mg+12.5 mg Tablet combines Losartan Potassium and Hydrochlorothiazide, manufactured by Square Pharmaceuticals PLC. It is used for the treatment of hypertension.
Indications
Angilock Plus is indicated for the treatment of hypertension. It is also indicated to reduce the risk of stroke in patients with hypertension and left ventricular hypertrophy.
Pharmacology
Losartan selectively blocks the binding of angiotensin II to the AT1 receptor, blocking its vasoconstrictor and aldosterone-secreting effects. Hydrochlorothiazide is a thiazide diuretic that increases excretion of sodium and chloride, reducing plasma volume and lowering blood pressure. The combination provides complementary blood pressure control.
Dosage and Administration
The usual starting dose of the 50/12.5 combination is one tablet once daily. If blood pressure is not adequately controlled, the dose may be increased to 100/25 once daily. The antihypertensive effect is generally attained within three weeks of starting therapy. May be administered with or without food, and with other antihypertensive agents.
Interaction
Concomitant use of potassium-sparing diuretics, potassium supplements, or salt substitutes containing potassium may increase serum potassium. NSAIDs such as indomethacin may blunt the antihypertensive effect. Alcohol, barbiturates or narcotics may potentiate orthostatic hypotension. Dosage adjustment of antidiabetic drugs may be required.
Contraindications
Contraindicated in patients hypersensitive to any component of this product, and in patients with anuria or hypersensitivity to other sulfonamide-derived drugs due to the Hydrochlorothiazide component.
Side Effects
Side effects are usually mild and include symptomatic hypotension, dizziness, vertigo, headache, and occasional hyperkalaemia. Less commonly: gastrointestinal disturbances, palpitation, oedema, rash and sleep disorders.
Pregnancy and Lactation
Angiotensin-II receptor antagonists should be avoided in pregnancy unless essential, as they may adversely affect fetal and neonatal blood pressure control and renal function. Not recommended during breastfeeding.
Precautions and Warnings
Monitor serum electrolytes periodically. Hypokalemia may rarely develop, especially with brisk diuresis or severe cirrhosis. Use with caution in impaired renal function and symptomatic hypotension.
