Canaglif 100 mg Tablet

Overview

Canaglif 100 mg Tablet contains Canagliflozin Hemihydrate, manufactured by Square Pharmaceuticals PLC. It is an oral sodium-glucose co-transporter 2 (SGLT2) inhibitor used to improve glycaemic control in adults with type 2 diabetes mellitus. It also reduces the risk of major cardiovascular events in adults with type 2 diabetes and established cardiovascular disease, and slows the progression of diabetic kidney disease.

Dosage and Administration

The usual starting dose is 100 mg once daily, taken before the first meal of the day. The dose may be increased to 300 mg once daily if additional glycaemic control is needed and the patient tolerates the lower dose, as advised by the physician. Renal function should be assessed before initiating and monitored periodically.

Interaction

Use with insulin or insulin secretagogues may increase the risk of hypoglycaemia; dose reduction of the insulin or secretagogue may be needed. Rifampicin and other UGT enzyme inducers may reduce canagliflozin exposure. Digoxin levels may be increased with concomitant use.

Contraindications

Contraindicated in type 1 diabetes, diabetic ketoacidosis, severe renal impairment (eGFR below 30 ml/min/1.73 m²), end-stage renal disease on dialysis, and known hypersensitivity to canagliflozin.

Side Effects

Genital mycotic infections, urinary tract infections, polyuria, volume depletion, and increased urination. Rare but serious risks include diabetic ketoacidosis (even at normal blood glucose), lower limb amputations, and bone fractures with long-term use.

Pregnancy and Lactation

Not recommended during pregnancy or breastfeeding due to potential harm to the developing kidney; consult a physician for safe alternatives in these situations.

Precautions and Warnings

Monitor renal function before and during therapy. Ensure adequate hydration. Interrupt therapy before major surgery or in situations of prolonged fasting. Patients with a history of lower limb problems should be monitored carefully. Patients should report any symptoms of ketoacidosis (nausea, vomiting, abdominal pain, breathlessness) even when blood glucose appears normal.
