Overview
Cilosta 100 mg Tablet contains Cilostazol, manufactured by Square Pharmaceuticals PLC. It is used to reduce the symptoms of intermittent claudication by improving walking distance.
Indications
Cilosta is indicated for the reduction of symptoms of intermittent claudication, as indicated by an increased walking distance. Intermittent claudication, also known as vascular claudication, is a symptom that describes muscle pain (pain, cramps, numbness, or a feeling of fatigue) during light exertion.
Pharmacology
Cilostazol is a quinolinone derivative that specifically inhibits cellular phosphodiesterase III (PDE III) and suppresses cAMP degradation, with a resultant increase in cAMP in platelets and blood vessels, leading to inhibition of platelet aggregation and vasodilation.
Dosage and Administration
The recommended dosage of Cilostazol is 100 mg twice daily, taken at least half an hour before or two hours after breakfast and dinner. A dose of 50 mg twice daily should be considered during coadministration of Fluconazole, Ketoconazole, Itraconazole, Erythromycin, Clarithromycin, Fluvoxamine, Fluoxetine, Nefazodone, Sertraline and Diltiazem. Safety and effectiveness in pediatric patients have not been established.
Interaction
Omeprazole and Erythromycin significantly increase the systemic exposure of Cilostazol and/or its major metabolites. Higher concentrations have been observed with concurrent Diltiazem use.
Contraindications
Contraindicated in patients with congestive heart failure of any severity, and in patients with known or suspected hypersensitivity to any of its components.
Side Effects
The most common side effects are headache, diarrhoea, vomiting and leg cramps. Less frequent side effects include anorexia and oedema.
Pregnancy and Lactation
There are no adequate and well-controlled studies in pregnant women. Transfer of Cilostazol into milk has been reported in animal studies; a decision should be made to discontinue nursing or discontinue the drug.
Precautions and Warnings
Use with caution in patients with any degree of heart failure. There is no information regarding the efficacy or safety of concurrent use with Clopidogrel.
