Indications

Baloxa is an influenza virus polymerase acidic (PA) endonuclease inhibitor indicated for: * Treatment of acute uncomplicated influenza in patients 12 years of age and older who have been symptomatic for no more than 48 hours and who are otherwise healthy or at high risk of developing influenza-related complications * Post-exposure prophylaxis (PEP) of influenza in patients 12 years of age and older following contact with an individual who has influenza

Pharmacology

Baloxavir Marboxil is an antiviral polymerase endonuclease inhibitor prodrug that is converted by hydrolysis to Baloxavir. Baloxavir inhibits the endonuclease activity of the polymerase acidic (PA) protein, an influenza virus-specific enzyme in the viral RNA polymerase complex required for viral gene transcription, resulting in inhibition of influenza virus replication.

Dosage & Administration

Take a single dose orally within 48 hours of symptom onset with or without food. Dosing by weight: 40 to less than 80 kg: two 20 mg tablets as a single dose (total 40 mg). At least 80 kg: two 40 mg tablets as a single dose (total 80 mg). For post-exposure prophylaxis, a single dose within 48 hours of exposure. Safety and effectiveness established in patients 12 years of age and older weighing at least 40 kg.

Side Effects

Adverse events (at least 1%) included diarrhea (3%), bronchitis (3%), nausea (2%), sinusitis (2%), and headache (1%).

Precautions & Warnings

Hypersensitivity such as anaphylaxis, angioedema, urticaria, and erythema multiforme may occur; initiate appropriate treatment if an allergic-like reaction occurs. Serious bacterial infections may begin with influenza-like symptoms; Baloxa has not been shown to prevent such complications.

Contraindications

Contraindicated in patients with a history of hypersensitivity to Baloxavir Marboxil or any of its ingredients.

Pregnancy & Lactation

There are no adequate and well-controlled studies in pregnant women. There are no data on the presence of baloxavir marboxil in human milk.
