Comprid 30 mg XR Tablet contains Gliclazide (Modified Release), a second-generation sulfonylurea antidiabetic agent manufactured by Square Pharmaceuticals PLC. It stimulates insulin secretion from pancreatic beta cells and is used in the management of type 2 diabetes mellitus in adults when diet and exercise alone are insufficient.
Dosage and Administration
Adults: 30 mg once daily with breakfast; may be increased to a maximum of 120 mg/day in divided doses as needed, as prescribed.
Interaction
Risk of hypoglycaemia increased with other antidiabetic drugs, alcohol, NSAIDs, sulfonamides, and fibrates. Rifampicin and barbiturates may reduce efficacy. Fluconazole and other CYP2C9 inhibitors may increase gliclazide levels.
Contraindications
Contraindicated in type 1 diabetes, diabetic ketoacidosis, severe renal/hepatic impairment, hypersensitivity to sulfonylureas, and during pregnancy and breastfeeding.
Side Effects
Hypoglycaemia is the most important adverse effect. GI disturbances, weight gain, and rarely haematological reactions may occur.
Pregnancy and Lactation
Contraindicated in pregnancy; insulin is preferred. Avoid during breastfeeding.
