Overview

Depram 25 mg Tablet contains Imipramine Hydrochloride, a tricyclic antidepressant (TCA) manufactured by Square Pharmaceuticals PLC. It inhibits reuptake of norepinephrine and serotonin, elevating mood. It is used for the treatment of major depressive disorder, nocturnal enuresis (bedwetting) in children, panic disorder, and chronic pain syndromes.

Dosage and Administration

Adults (depression): initially 75 mg/day in divided doses, gradually increased to 150–300 mg/day as needed; elderly patients start at lower doses. Nocturnal enuresis in children: 25 mg at bedtime, up to 50 mg for older children. Take with or after food.

Interaction

Serious interaction with MAO inhibitors (risk of hypertensive crisis — avoid concurrent use and allow 14-day washout). Increases CNS depression with alcohol and sedatives. Anticholinergic effects additive with other antimuscarinic drugs. CYP2D6 inhibitors increase imipramine levels.

Contraindications

Contraindicated within 14 days of MAOI use, in acute myocardial infarction, known hypersensitivity to TCAs, arrhythmias, severe liver disease, and angle-closure glaucoma.

Side Effects

Dry mouth, sedation, constipation, urinary retention, blurred vision, weight gain, orthostatic hypotension, and tachycardia. Overdose can cause fatal cardiac arrhythmias.

Pregnancy and Lactation

Potential teratogen; avoid in first trimester. Use with caution later in pregnancy under physician supervision. Passes into breast milk; not recommended during breastfeeding.

Precautions and Warnings

Monitor for suicidal ideation, especially at treatment initiation and dose changes. Do not stop abruptly (taper gradually). Avoid alcohol. ECG monitoring advisable in cardiac patients and elderly.

Storage Conditions

Store below 30°C in a cool, dry place, protected from light. Keep out of the reach of children.
