Overview
Doxacil 100 mg Capsule contains Doxycycline Hydrochloride, a broad-spectrum second-generation tetracycline antibiotic manufactured by Square Pharmaceuticals PLC. Doxycycline inhibits bacterial protein synthesis by binding to the 30S ribosomal subunit. It is effective against a wide range of Gram-positive and Gram-negative bacteria, atypical organisms, and intracellular pathogens. Clinical uses include respiratory tract infections, urinary tract infections, chlamydia and other sexually transmitted infections, acne vulgaris, malaria prophylaxis and treatment, Lyme disease, and rickettsia and mycoplasma infections.
Dosage and Administration
Adults: 100 mg twice daily on day one, then 100 mg once daily; or 100 mg twice daily throughout depending on severity and indication. Take with food and a full glass of water to avoid oesophageal irritation. Malaria prophylaxis: 100 mg daily starting 1–2 days before travel, continuing during and for 4 weeks after.
Interaction
Antacids, calcium, iron, and dairy products reduce doxycycline absorption — take doxycycline 2 hours before or after these. May reduce efficacy of oral contraceptives. Rifampicin, phenytoin, and carbamazepine reduce levels. Potentiates warfarin effect.
Contraindications
Contraindicated in known hypersensitivity to tetracyclines, pregnancy (risk of permanent tooth staining), children under 8 years, and severe hepatic impairment.
Side Effects
Nausea, vomiting, diarrhoea, oesophageal irritation, photosensitivity (avoid prolonged sun exposure), and headache. Dental/bone discolouration if used in young children or during pregnancy.
Pregnancy and Lactation
Contraindicated in pregnancy — causes permanent discolouration of developing teeth and may affect foetal bone development. Excreted in breast milk — avoid during breastfeeding.
Precautions and Warnings
Take with food and adequate water; sit upright for 30 minutes after taking. Avoid prolonged sun exposure — use sunscreen. Complete the full course. Do not take with dairy products, antacids, or iron supplements.
