Overview

GOL Oral Solution contains Polyethylene Glycol 3350 (Macrogol) combined with electrolytes — Sodium Chloride, Sodium Bicarbonate, and Potassium Chloride. Each 25 ml concentrated oral solution provides Polyethylene Glycol 3350 13.125 gm, Sodium Chloride 350.7 mg, Sodium Bicarbonate 178.5 mg, and Potassium Chloride 46.6 mg. Macrogol exerts an osmotic action in the gut that induces a laxative effect, increasing stool volume and triggering colon motility via neuromuscular pathways. The combined electrolytes are exchanged across the intestinal mucosa with serum electrolytes without net gain or loss of sodium, potassium, or water. GOL is indicated for adults and children over 12 years for effective relief from constipation, treatment of chronic constipation, and resolution of fecal impaction.

Dosage and Administration

Constipation (adults and children aged 12 years and above): 25 ml of GOL Oral Solution added to 100 ml of water once daily (total volume 125 ml). May be increased to 2–3 doses of 25 ml daily, each added to 100 ml of water, according to individual response.

Fecal Impaction: 8 doses of 25 ml daily, each added to 100 ml of water. A course of treatment for fecal impaction does not normally exceed 3 days.

Direction of use: 25 ml of oral solution added to 100 ml of water. Any unused diluted solution should be discarded within 24 hours. Patients with renal insufficiency require no dosage adjustment.

Interaction

There is a possibility that the absorption of other medicinal products could be transiently reduced during use with GOL. Isolated reports of decreased efficacy have been observed with some concomitantly administered medicinal products, particularly anti-epileptics. Patients taking medications with narrow therapeutic windows should consult their physician.

Contraindications

GOL Oral Solution is contraindicated in patients with intestinal perforation or obstruction due to structural or functional disorder of the gut wall, ileus, severe inflammatory conditions of the intestinal tract (such as Crohn's disease, ulcerative colitis, and toxic megacolon), and known hypersensitivity to the active substances or any excipient.

Side Effects

GOL is generally well tolerated. Reported side effects include allergic reactions, electrolyte disturbances (particularly hyperkalaemia and hypokalaemia), abdominal pain, diarrhoea, headache, and peripheral oedema. Discontinue if symptoms of fluid or electrolyte imbalance occur.

Pregnancy and Lactation

No clinically significant effects during pregnancy are anticipated, as systemic exposure to Macrogol 3350 is negligible. GOL Oral Solution can be used during pregnancy. No effects on the breastfed newborn or infant are anticipated since the systemic exposure of the breastfeeding woman to Macrogol 3350 is negligible. GOL can be used during breastfeeding.

Precautions and Warnings

In patients with impaired cardiovascular function, for the treatment of fecal impaction, the dose should be divided so that no more than two doses are taken within any one hour. If patients develop symptoms indicating fluid or electrolyte shifts — such as oedema, shortness of breath, increasing fatigue, dehydration, or cardiac failure — GOL should be stopped immediately and the abnormality treated appropriately. Prolonged use of laxatives is undesirable and may lead to dependence. In case of overdose, severe pain or distension can be treated by nasogastric aspiration; extensive fluid loss may require correction of electrolyte disturbances.
