Lesovir-C combines ledipasvir (NS5A replication complex inhibitor) and sofosbuvir (NS5B polymerase inhibitor nucleotide analog prodrug). The dual mechanism targets different stages of HCV replication, providing potent pangenotypic antiviral activity with high barrier to resistance.

Indications

  • Chronic Hepatitis C virus (HCV) genotype 1, 4, 5, and 6 infection in adults
  • Treatment-naive and treatment-experienced chronic HCV patients
  • HCV patients with or without cirrhosis
  • HCV/HIV co-infected patients

Dosage & Administration

Adults: 1 tablet (Ledipasvir 90mg/Sofosbuvir 400mg) once daily with or without food for 8, 12, or 24 weeks depending on genotype, prior treatment history, and degree of liver fibrosis. Specialist hepatologist prescription required.

Contraindications & Precautions

  • Hypersensitivity to ledipasvir, sofosbuvir, or excipients
  • Co-administration with rifampicin, rifabutin (reduce sofosbuvir levels)
  • Severe renal impairment (GFR <30 mL/min) — co-administration with amiodarone contraindicated
  • Pregnancy: use only if clearly needed

Side Effects

Common: fatigue, headache, nausea, diarrhea, insomnia. Bradycardia reported when co-administered with amiodarone.

Drug Interactions

Amiodarone: serious symptomatic bradycardia — contraindicated. Rifampicin, St. John's Wort: reduce ledipasvir/sofosbuvir levels. Antacids containing aluminium/magnesium: separate by 4 hours.
