Zomitan 2.5 mg Tablet

What is Zomitan?

Zomitan contains Zolmitriptan, a selective serotonin (5-HT1B/1D) receptor agonist used for the acute treatment of migraine headaches with or without aura in adults. It works by causing constriction of cranial blood vessels and inhibiting the release of pro-inflammatory neuropeptides that contribute to migraine pain.

Indications

Zomitan is indicated for the acute treatment of migraines with or without aura in adults. It is not intended for the prevention of migraines or for use in cluster headaches.

Pharmacology

Migraines are likely due to local cranial vasodilatation and/or the release of sensory neuropeptides through nerve endings in the trigeminal system. The therapeutic activity of Zolmitriptan in migraine treatment is attributed to its agonist effects at 5-HT1B/1D receptors on intracranial blood vessels and sensory nerves of the trigeminal system, resulting in cranial vessel constriction and inhibition of pro-inflammatory neuropeptide release. Zolmitriptan is rapidly and well absorbed after oral administration.

Dosage & Administration

The recommended starting dose is 2.5 mg tablet. The maximum recommended single dose is 5 mg. If the migraine has not resolved by 2 hours after the first dose, or returns after transient improvement, a second dose may be taken at least 2 hours after the first. The maximum daily dose is 10 mg within any 24-hour period.

Interaction

Caution is advised when Zolmitriptan is co-administered with CYP1A2 inhibitors (e.g., cimetidine, fluvoxamine, quinolones), other triptans, ergotamine-containing medications, or MAO inhibitors, as these may increase the risk of serotonin syndrome or excessive vasoconstriction. Concomitant use with other 5-HT1 agonists or ergotamine derivatives within 24 hours should be avoided.

Contraindications

Contraindicated in patients with ischemic coronary artery disease, history of stroke or transient ischemic attack, peripheral vascular disease, uncontrolled hypertension, concurrent or recent (within 2 weeks) use of a monoamine oxidase inhibitor, and in patients with known hypersensitivity to Zolmitriptan.

Side Effects

Most common adverse reactions include neck/throat/jaw pain or pressure, dizziness, paresthesia, asthenia, somnolence, sensations of warmth or cold, nausea, a feeling of heaviness, and dry mouth.

Pregnancy & Lactation

Pregnancy Category C. Zolmitriptan should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is not known whether Zolmitriptan is excreted in human milk; a decision should be made whether to discontinue nursing or discontinue the drug, taking into account the importance of the drug to the mother.

Precautions & Warnings

There have been rare reports of serious cardiac adverse events, including acute myocardial infarction, occurring within a few hours of administration. Sensations of tightness, pain, and pressure may occur in the chest, throat, neck, and jaw after treatment. Not recommended for use in patients under 18 years of age. Should be used with caution in patients with risk factors for coronary artery disease.
