Overview

Apsol Oral Paste 5% contains Amlexanox and is used for the treatment of aphthous ulcers (mouth ulcers/canker sores).

Mechanism of Action

The exact mechanism by which Amlexanox accelerates healing of aphthous ulcers is not fully established. In vitro, it is a potent inhibitor of the formation and release of inflammatory mediators (histamine and leukotrienes) from mast cells, neutrophils, and mononuclear cells, and has shown anti-allergic and anti-inflammatory activity in animal studies.

Indications

Apsol is indicated for the treatment of aphthous ulcers.

Dosage and Administration

Apply the paste as soon as possible after noticing ulcer symptoms, and continue four times daily, preferably after oral hygiene following breakfast, lunch, dinner, and at bedtime. Dry the ulcer gently with a soft, clean cloth and wash hands before application. Squeeze about ¼ inch (0.5 cm) of paste onto a moistened fingertip and gently dab it onto the ulcer; repeat for additional ulcers. Wash hands after application, and flush eyes promptly with water if accidental contact occurs. Continue use until the ulcer heals; if significant healing or pain relief has not occurred within 10 days, consult a physician.

Contraindications

Contraindicated in patients with known hypersensitivity to Amlexanox or other ingredients in the formulation.

Side Effects

Transient pain, stinging, and/or burning at the application site have been reported in 1–2% of patients. Less frequent (<1%) reactions include contact mucositis, nausea, and diarrhea.

Pregnancy and Lactation

US FDA pregnancy category B. Animal studies have not shown adverse fetal effects, but adequate controlled studies in pregnant women are lacking; use during pregnancy only if clearly needed. Amlexanox has been found in the milk of lactating animals, so caution is advised when used by a nursing woman.

Special Precautions

Wash hands immediately after applying the paste directly to ulcers with the fingertips. Discontinue use if a rash or contact mucositis occurs. Safety and effectiveness in pediatric patients have not been established.

Store in a cool and dry place. Protect from light. Keep out of reach of children.
