DESCRIPTION

A-FLOX (Flucloxacillin) is a bactericidal antibiotic with considerable activity against the following Gram-positive organisms: penicillinase producing Staphylococcus aureus, penicillin sensitive Staphylococcus aureus, Beta-haemolytic Streptococci (Streptococcus pyogenes), Diplococcus pneumoniae etc. It is not active against Gram-negative bacilli, methicillin resistant Staphylococcus aureus, Streptococcus faecalis. Flucloxacillin is absorbed about two times than Cloxacillin from the gastrointestinal tract. Absorption is reduced by the presence of food in the stomach. Flucloxacillin kills bacteria by inhibiting the synthesis of the bacterial cell wall.

COMPOSITION

Capsule 250 : Each capsule contains Flucloxacillin Sodium BP equivalent to 250 mg Flucloxacillin.

Capsule 500 : Each capsule contains Flucloxacillin Sodium BP equivalent to 500 mg Flucloxacillin.

Powder for suspension : Each 5 ml reconstituted suspension contains Flucloxacillin Sodium BP equivalent to 125 mg Flucloxacillin.

Injection 250 : Each vial contains Flucloxacillin Sodium BP equivalent to 250 mg Flucloxacillin.

Injection 500 : Each vial contains Flucloxacillin Sodium BP equivalent to 500 mg Flucloxacillin.

INDICATIONS

A-FLOX is indicated for the treatment of infections caused by Grampositive microorganisms. It is also indicated for the treatment of infections caused by penicillinase producing microorg-anisms. These type of infections include: Skin and soft tissue infections : Abscess, carbuncle, furunculosis, cellulitis, infected wound and burn and other skin infections. Respiratory tract and E.N.T. infections : Pneumonia, lung abscess, empyema, sinusitis, pharyngitis, tonsillitis, otitis media and externa. Other infections : Osteomyelitis, meningitis, enteritis, endocarditis, urinary tract infection, septicaemia.

DOSAGE AND ADMINISTRATION

The oral dose should be administered ½ -1 hour before meals.

Oral :

Adults : Usual adult dose is 250 mg every 6 hours or two measuring spoonful of suspension every 6 hours. The dose may be doubled in severe cases.

Children (2-10 years) : ½ of the adult dose; (1 measuring spoonful 6 hourly).

Children (under 2 years) : ¼ of the adult dose. In severe infections, the dosage may be increased.

Injection :

Adults : Usually 250 mg IM four times daily. Can also be administered intra-venously in a dose of 250 mg to 1 g four times daily by slow injection over 3 to 4 minutes or by intravenous infusion.

The dose may be doubled in severe cases. In case of intravenous infusion, powder can be dissolved in any type of fluid except blood and proteinaceous fluid.

Children : Children up to 2 years of age may be given one quarter of the adult dose and those aged 2-10 years, one half of the adult dose.

SIDE EFFECTS

Nausea, vomiting, diarrhea may occur. These effects are mild and rare. Allergic reactions may occur in hypersensitive patients. In such cases treatment should be discontinued.

CONTRAINDICATIONS

Flucloxacillin can not be given to Penicillin or Cephalosporin hypersensitive patients.

PRECAUTIONS

No adverse reaction has been reported on embryo. However, the use of Flucloxacillin in pregnancy should be reserved for cases considered essential by the physician.

SUPPLY

Capsule 250 : Each box contains 12 X 4 capsules in strips.

Capsule 500 : Each box contains 12 X 4 capsules in strips.

Powder for suspension : Each bottle contains required amount of

powder to reconstitute 100 ml suspension. Injection 250 : Each box

contains five vials of 250 mg Flucloxacillin powder for injection and

five ampoules of 5 ml water for injection.

Injection 500 : Each box contains five vials of 500 mg Flucloxacillin

powder for injection and five ampoules of 5 ml water for injection.
